Description

This assay employs a two-site sandwich ELISA to quantitate YWHAE in samples. An antibody specific for YWHAE has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any YWHAE present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for YWHAE is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of YWHAE bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: YWHAeproduct belongs to the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals, and this protein is 100% identical to the mouse ortholog. It interacts with CDC25 phosphatases, RAF1 and IRS1 proteins, suggesting its role in diverse biochemical activities related to signal transduction, such as cell division and regulation of insulin sensitivity. It has also been implicated in the pathogenesis of small cell lung cancer.Two transcript variants, one protein-coding and the other non-protein-coding, have been found for this gene.
